(1998) Genetics of psoriasis: paternal inheritance and a locus on chromosome 6p

Since 1994, three main genetic loci (on chromosomes 17q, 4q, and 6p) have been reported in genome scans. With a view to elucidating the genetic basis of psoriasis, we have carried out linkage analysis in a large number of families with well-characterized psoriasis. Several genetic loci have been reported for psoriasis, but none has been specifically linked to psoriatic arthritis (PsA), a condition that affects 10 of patients with psoriasis. Genetics of psoriasis: paternal inheritance and a locus on chromosome 6p. Psoriasis and psoriatic arthritis (PsA) are clearly complex genetic disorders that result from an interplay between multiple genetic and environmental factors. Hum Mol Genet1998;7:1599609. Genome-wide linkage scans have identified multiple loci linked to each disease and revealed overlap with psoriasis and atopic dermatitis susceptibility loci on chromosomes 1q21, 3q21, 17q25 and 20p12. In each of the cases described above, psoriasis is inherited as an autosomal dominant trait with reduced penetrance, and the two-point LOD scores exceed the conventionally accepted threshold of 3. These have consistently identified a limited number of regions containing genes influencing asthma, including chromosomes 2, 4, 5, 6, 7, 11, 12, 13 and 16 (74,83). Bhalerao, J. and Bowcock, A.M. (1994) showed in a population genetic study that a recessive mode of inheritance was compatible with distribution among first-degree relatives, given a high gene frequency. (1998) use the standard nomenclature for the psoriasis susceptibility locus on chromosome 6, PSORS1. The evidence for linkage in sib-pair analysis was greatest when the allele was of paternal origin and was most significant in those families without psoriatic arthritis.

Another explanation is that psoriasis and PsA may share susceptibility factors, which may be genetic, environmental or both. Multiple studies indicate genetic linkage of psoriasis to a locus on chromosome 6p21.3 (PSORS1), accounting for 35 to 50 of the genetic contribution to psoriasis (Burden et al, 1998; Nair et al, 1997; Trembath et al, 1997). Genetics Of Psoriasis: Paternal Inheritance And A Locus On Chromosome 6p

Since the PSORS1 locus within the MHC region on 6p provides the strongest linkage with psoriasis in genome-wide linkage scans, candidate genes in this region have been investigated.
